On April 19, 1943, Swiss chemist Albert Hofmann embarked on a bicycle ride that would alter the course of scientific history. After intentionally ingesting 250 micrograms of lysergic acid diethylamide (LSD)—a compound he had synthesized five years earlier—Hofmann cycled home from his laboratory at Sandoz Pharmaceuticals in Basel, Switzerland. What followed was humanity’s first documented, intentional LSD-induced psychedelic experience, marked by vivid hallucinations, distorted perceptions of time, and profound introspection. This day, now celebrated as Bicycle Day, marked the dawn of psychedelic neuroscience and catalyzed a paradigm shift in our understanding of consciousness, mental health, and the brain’s hidden potential.
Hofmann’s accidental discovery of LSD’s psychoactive properties in 1943 (during earlier lab work, he absorbed a trace dose through his fingertips) and his subsequent intentional experiment revealed a molecule with unparalleled power to illuminate the mind’s inner workings. LSD’s ability to induce altered states of consciousness—while leaving the user physically unharmed at controlled doses—made it a revolutionary tool for researchers.
LSD’s structural similarity to serotonin (a neurotransmitter regulating mood, cognition, and perception) allowed scientists to trace its binding to serotonin receptors, particularly the 5-HT2A receptor. This discovery unlocked critical insights into how neurotransmitters shape consciousness, paving the way for modern antidepressants like SSRIs.
Modern fMRI studies show that psychedelics like LSD temporarily quiet the DMN—a brain network linked to self-referential thought, ego, and rumination. This suppression correlates with ego dissolution and heightened interconnected thinking, offering clues to treating depression, anxiety, and addiction by “resetting” maladaptive neural pathways.
